Using instruction

Ampicillin- 500 mg.

capsules

Description:

Firm white colour gelatinous capsules.

Composition of one capsule:

Active substance:

Ampicillin trihydrate - 500 mg.

Aiding substances: starch; calcium stearate or magnesium stearate.

Pharmacological operation:

Antibacterial antibiotic action from penicillin group. It is active in the relation with gram-positive (alpha and beta-haemolytic streptococci, Streptococcus pneumoniae, Staphylocjccus spp., Bacillus antheacis, Clostridium spp., the majority of enterococcus) and gram-negative (Haemophilus infuenzae, Neisseria meningitides, Proteus mirabilis, Escherichia coli, many kinds of salmonella and shigell) microorganisms.

Pharmacodynamics:

Pharmacological mechanism basis of bactericidal action of ampicillin is its ability to carry out inhibition of biosynthesis of walls of cells of the pathogenic microorganisms which are in a growth phase. It carries out inhibition of peptidoglycan and transpeptidaze polymers, interferes formation of peptide union in the synthesis process of microorganism's cell of wall. The formed defects of cover reduces osmotic stability of bacterial cells and cause their lysis.

Pharmacokinetics: Ampicillin is rather steady in acid area of digestive juices and, as a rule, at peroral reception is well absorbed from gastrointestinal tract. 20 % of ampicillin relates to proteins of blood plasma in the course of circulation. The maximum concentration in blood is reached in 1,5-2 hours after intake. Approximately 30 % from oral intake dose are allocated with urine during 6 - 8 hours period. High concentration of ampicillin is reached in the bubble. Insignificant diffusion in a spinal cord liquid, except meningitis cases when is marked  the reach of  high  concentration of ampicillin.

Indication

  • Infections of the upper and inferior respiratory tract (pneumonia, pulmonary abscess, bronchitis, sinusitis, pharyngitis, otitis);
  • Infections of kidneys and urinogenital systems (cystitis, pyelonephritis, gonorrhoea);
  • Infections of gastroenteric system ( peritonitis, enterocolitis, cholecystitis, typhoid);
  • Infections of musculoskeletal system, meningitis.

Contra-indication

Heightened sensibility toward penicillin and cephalosporin. Allergic diathesis,  bronchial asthma, infectious mononucleosis, lymphatic leukemia.

Side-effects:

Nausea, vomiting, diarrhea, pains in the field of anus, stomatitis, irritability,  sleeplessness, dizziness, difficulty of breath,  tachycardia, pains in joints,  dysbacteriosis,  candidiasis in oral or vaginas cavities; hemorrhagic colitis, allergic reactions, dermatitis, exudative erythema, itch,  rash.

Order of reception and dosage:

The preparation dose depends on sensitivity of the microbe which has caused illness, to ampicillin and difficulty of infectious process. Usually, for adults and children over 16 years prescribed 500-700 mg 2 times in a day or 375-500 mg 3 times in a day.

For children from 3 till 10 years - 375 mg 2 times or 250 mg 3 times a day.

For children till 1 year - 125 mg 2 times or 100 mg 3 times in a day.

The daily dose for children makes 30 mg/kg (to 60 mg/kg), in case of acute gonorrhoea simultaneous reception of 3 g preparation in combination with 1 g probenecid is supposed.

Interaction with other medical preparations:

Ampicillin can cause strengthening of peroral anticoagulants operation taking simultaneously.

Simultaneous reception with probenecid, concentration of ampicillin raises in blood plasma.

At simultaneous reception with chloroquin absorbability of ampicillin can decrease.

Ampicillin reduces efficiency of oestrogen containing contraceptives perorally.

Special instructions:

The pregnancy and lactation period: At pregnancy Ampicillin  may be taken only when the expected advantage for mother exceeds potential risk for the child;

Ampicillin gets to mother's milk and consequently it is necessary to interrupt chest feeding during treatment.

Influence on ability of management of vehicles and mechanisms:

Ampicillin does not operate on ability of driving vehicles and mechanisms.

Overdose:

Symptoms: Allergic reactions, dyspepsic events.

Treatment: It is necessary to stop preparation reception, cause of vomiting, wash out of stomach. After that symptomatic treatment is necessary. Excretion of Ampicillin from blood circulation system is possible also by hemodialysis.
